VALIDATED CHROMATOGRAPHIC METHOD FOR THE DETERMINATION OF OLEANOLIC ACID IN ACHRANTHES ASPERA LINN

Achyranthes aspera Linn (Fam: Amaranthaceae) is widely used plant in Indian medicine for the treatment of various ailments. Among the complex constituents of this plant, oleanolic acid, a triterpene acid is reported in the seeds and roots of A.aspera. A simple sensitive and precise HPTLC method for the quantitative analysis of oleanolic acid in seeds of A.aspera was developed. Chromatographic separation of oleanolic acid was performed on silica gel 60 F254 plates using hexane-ethylacetate-acetic acid (8+4+0.2v/v/v) as mobile phase. The plates were derivatized using anisaldehyde-sulphuric acid reagent and scanned densitometrically at 530 nm. The method was val idated for precision, repeatability and accuracy. A polynomial relation was obtained for oleanolic acid in the range of 200 -1200ng/spot. The method was found to be precise, with relative standard deviation (RSD) values for intraday analyses in the range of 1.02-3.71%. The method was further applied to study the effect of germination of seeds of A.aspera on the content of oleanolic acid. The content of oleanolic acid in the non-germinated and germinated seeds of A.aspera was found to be 0.343 ± 0.0071w/w and 0.625 ± 0.018%w/w respectively. The proposed method is rapid, simple and accurate and can be used for the routine quality control of oleanolic acid
